Nandrolone (Deca Durabolin) Nandrolone is one of the most commonly used steroids for muscle growth, and most of the research into its potential in the treatment of muscular dystrophy has been conducted on non-human primates (Kelton et al., 1989; Gee et al., 1994). However, for some unknown reason, humans have difficulty synthesising and maintaining sufficient amounts of Nandrolone in muscle tissues. In a study of normal men, there was insufficient Nandrolone in muscle to stimulate increases in the total protein content over the 4-5 wk of the study (Kamil et al., 1994). In addition to the lack of Nandrolone in human muscle, it has been found that it appears to be deficient in several other tissues of the body. This deficiency has been found to be related to the enzyme CYP3A4, and other known deficiencies are the deficiency in CYP2C19, CYP2C9, and CYP2C19A1 as well as CYP3A4 (Kilpatrick, 1999). Several studies have shown that the amount of Nandrolone that is required to produce an increase in total protein levels over the course of 3 wk of treatment with testosterone is about 10-50% lower in man compared to women (Kilpatrick, 1999). The differences in the amounts required may be partly due to sex steroid hormone receptors and their interactions with the enzymes of the synthesis pathway leading to the formation of muscle. This pathway is dependent on a number of proteins for synthesis of a wide array of different proteins (Chen et al., 1994). It appears that an increased number of steroid receptor-deficient enzymes (as in the case at levels of 14,000-39,000), or reduced enzymes in response to increases in concentrations of Nandrolone (as in the case of 14,000-39,000), will limit the ability to activate the proper enzymes and thus will result in an inadequate dose to activate the appropriate proteins. Many of the enzymes involved in this pathway are believed to be in an essential role in maintaining normal protein synthesis as compared to the case of an enzyme deficient due to the presence of CYP2C19. The most likely reason that a deficiency exists at these high levels is that Nandrolone is required to function as a coenzyme for the synthesis of amino acids and thus to help convert the amino acids in the muscle tissue into the building blocks of proteins which can be used by the body.
